MRI should be the most sensitive way to determine whether your father has a brain tumor. MRI can be performed with and without intravenous contrast and sometimes pathology is seen only with the contrast. You didn't mention the time period between the MRI scans. Was the MRI scan that was negative performed earlier than the one that was positive? It is possible that initially the pathology was too small to be seen by MRI but perhaps it was identified later after it progressed.

Overall, MRI is the most sensitive study for brain tumor. You might want to have the radiologist who interpreted the second study offer an opinion.
